Hyaluronic Acid-Solid Lipid Nanotransporter Serum for Enhanced Topical Tretinoin Delivery: A Comprehensive Study on Skin Safety and Efficacy

Solid lipid nanotransporters (SLNs) have garnered attention as efficient carriers for drug delivery to the skin, enhancing permeability and stability. Hyaluronic acid (HA) is a well-known moisturizing agent with skin barrier-enhancing properties, making it a suitable candidate for combination with SLN to improve the efficacy of topical tretinoin.

Tretinoin, a retinoid widely used for acne and wrinkle treatment, faces limitations in delivery due to its instability and skin irritation potential. Researchers aimed to overcome these barriers by formulating a hyaluronic acid-based SLN (H-T-SLN) for stable, safe, and effective topical application.

Materials and Methods:
Hyaluronic acid-based SLNs were prepared by loading tretinoin into the lipid matrix. The formulation was evaluated for particle size, stability (6 months at 40°C), drug compatibility (DSC and XRD tests), and skin safety (hemolysis assay). A visual assessment of skin conditions, including wrinkle reduction, was conducted through a human study on participants using the H-T-SLN serum.

  1. Size and Stability: H-T-SLNs exhibited a superior particle size-to-volume ratio, aiding in enhanced skin penetration. Stability tests confirmed that the formulation remained stable for over 6 months at 40°C.
  2. Amorphous State Conversion: DSC and XRD analyses revealed that the formulation existed in an amorphous state, enhancing its bioavailability.
  3. Skin Safety: Hemolysis assays indicated compatibility with rat blood and no toxic effects, demonstrating its safety for skin application.
  4. Efficacy: Human trials showed a significant reduction in wrinkles after daily use, supporting the efficacy of H-T-SLN in anti-aging skincare. Long-term studies confirmed the immediate positive effects observed in short-term evaluations.


SLN nanotransporters represent a highly effective system for delivering tretinoin to the skin. The incorporation of hyaluronic acid further enhanced the penetration and stability of the formulation.
